Two-dimensional gel analysis demonstrates no structural alteration of HLA-B27 polypeptides between patients with ankylosing spondylitis and healthy individuals.

چکیده
After precipitation of the HLA-B27 antigen from the surface of peripheral blood lymphocytes (PBL) by means of an anti-HLA-B27 allospecific monoclonal antibody 2-dimensional gel electrophoresis was used to compare the structure of the B27 antigens derived from 5 patients with ankylosing spondylitis with that of healthy HLA-B27 positive counterparts. No significant difference in polypeptide structure was noted, which suggests that the pathogenesis of ankylosing spondylitis does not involve a structural alteration in cell surface HLA-B27 molecules.

Structural identity of human histocompatibility leukocyte antigen-B27 molecules from patients with ankylosing spondylitis and normal individuals.
Although the association between human histocompatibility leukocyte antigen (HLA) B27 and ankylosing spondylitis is the prototype of HLA-disease association, the mechanism underlying these associations has not been determined. We have investigated the possibility that the B27 molecules from patients with ankylosing spondylitis are different from those of normals, and only the "different" molecu...
متن کاملDistribution of HLA-B*27 Alleles in Pa-tients with Ankylosing Spondylitis in Iran
Background: HLA-B*27 is strongly associated with ankylosing spondylitis (AS). It represents a family of alleles that differ among ethnic groups. Objective: The aim of this study was to determine the distribution of HLA-B*27 alleles in AS patients and healthy controls in Isfahan (Iran). Methods: Sixty AS patients and 430 healthy blood donors were selected. All subjects were HLA-B*27 positive by ...
متن کاملASSOCIATION OF HLA-B27 WITH ANKYLOSING SPONDYLITIS IN ISFAHAN, IRAN
Using a standard microcytotoxicity (NIH) technique of tissue typing, the HLA-B27 antigen was identified in 30 out of 34 patients (HH.2%) with classical ankylosing spondylitis (AS), compared to 6 out of 70 controls (H.6%) (P < 0.005). We also found this antigen in 8 out of 76 (10.5%) patients with non-AS arthritis.
متن کاملAnkylosing spondylitis and HLA-B27: restriction fragment length polymorphism and sequencing of an HLA-B27 allele from a patient with ankylosing spondylitis.
Two groups of patients with ankylosing spondylitis (AS) from England and Poland were examined for restriction fragment length polymorphisms (RFLPs) associated with the disease. No preferential association was found between the 9.2 kb PvuII fragment in HLA-B27 positive patients with AS compared with HLA-B27 healthy subjects as had been previously reported. In the English group, however, a 14 kb ...
متن کاملHuman Leukocyte Antigen B27 in 453 Asian Indian Patients with Seronegative Spondyloarthropathy
Background: Spondyloarthropathies are a group of closely related inflammatory arthritis which involve the axial skeleton and are negative for rheumatoid factor. Objective: This case-control study was conducted to examine HLA- B27 positivity in patients with seronegative spondyloarthritis (SSA) as per ESSG criteria and compare the frequency with healthy controls because a lower positivity is rep...
